The objective of this study was to determine the effect of finasteride on the viability of MCF-7 cells treated with DHL-2-specific phosphodiesterase 5 inhibitor (PDE5i).
MDA-MB-231, a human breast cancer cell line, was exposed to DHL-2-specific phosphodiesterase 5 inhibitor (PDE5i), finasteride, for 48 h. DHL-2-specific phosphodiesterase 5 inhibitor (PDE5i) was added to the culture medium. DHL-2 was added to the culture medium in the presence of finasteride. The cells were exposed to finasteride, and the cell viability was assessed 24 h after treatment.
To test the effect of finasteride on cell viability and the effect of PDE5i, MCF-7 cells were cultured in high-glucose medium. Cells were then exposed to DHL-2-specific PDE5i. After DHL-2-specific PDE5i treatment, the cells were exposed to finasteride for 24 h.
Cells were exposed to DHL-2-specific PDE5i for 48 h. Then, the cells were harvested and lysed using trypsin. The protein levels of c-Fos, c-Myc, c-Jun and c-Myc were determined using Western blotting. The cells were lysed in ice for 30 min to remove the cell lysates. The cells were lysed using TRIzol reagent. The protein concentrations of the lysates were determined using the BCA Protein Assay (Beyotime, Japan). The protein amounts of c-Fos, c-Myc, c-Jun and c-Myc were quantified using the bicinchoninic acid protein assay (Thermo Fisher Scientific).
MDA-MB-231 cells were cultured in high-glucose medium with either finasteride (0.2 μM) or DHL-2-specific PDE5i (0.5 μM) for 24 h. Cells were then treated with finasteride, and the cell proliferation was determined using the Cell Counting Kit-8 (CK8) kit. The cells were harvested and lysed using TRIzol reagent. The cell lysates were suspended in ice for 10 min and centrifuged at 10,000 rpm for 5 min. The supernatants were collected, and the protein concentration of the supernatants was determined using the BCA protein assay kit (Thermo Fisher Scientific). The cell lysates were centrifuged again at 10,000 rpm for 5 min. The cell lysates were resuspended in ice and incubated for 1 h at 4 °C.
The cell viability was determined using the Cell Counting Kit-8 (CK8) kit (Thermo Fisher Scientific). The cells were lysed using a CCK8 substrate (Beyotime, Japan). The lysates were centrifuged at 10,000 rpm for 5 min. The supernatants were collected and the cell lysates were resuspended in ice for 1 h at 4 °C.
To detect the effect of finasteride on cell proliferation, MCF-7 cells were cultured in high-glucose medium with either finasteride (0.2 μM) or DHL-2-specific PDE5i (0.5 μM) for 24 h. Cells were then exposed to finasteride, and the cell proliferation was determined using the Cell Counting Kit-8 kit (Beyotime, Japan).
